How to count? With the Spiral method, a logarithmically decreasing volume of sample is dispensed on the surface of a rotating Petri dish in an Archimedes spiral. The volume is calibrated and known at every point of the Petri dish. Bacterial concentration is determined by dividing the number of colonies found by the volume dispensed in the same sector of the dish. For manual counting, count the colonies in the fi rst A or B outer segment and reach a minimum of 20 colonies. Repeat the same operation on the opposite side of the Petri dish. How does it work? 1. Questions What is special about the easyspiral Dilute? easyspiral Dilute is the only 2-in-1 equipment on the market making automatic serial dilutions and standardized plating. The accuracy of easyspiral Dilute is high: 0.5%. How can I check it? I usually dilute 1 ml to 1/10 and the easyspiral Dilute uses 0.5 ml to 1/10. Is easyspiral Dilute as precise with a volume divided by 2? It is easy and automatic. Test of the deposited volume during plating: Press on the plating volume test button. This function will fi ll a previously weighed beaker with 500 µl. Weigh the distributed volume to check the results. Test of the volume of the dilutions: Press on the dilution volumes test button. This function will fi ll 2 previously weighed beakers: one with 0.5 ml and the 2 nd one with 4.5 ml. Weigh the distributed volumes to check the results. Usually the average accuracy for the 9 ml tubes fi lling and 1 ml pipetting is 2% with the need to calibrate and regularly check the pipets you are using. easyspiral Dilute has an average accuracy of 0.5% on 0.5 ml and 4.5 ml volumes because it works with a high precision Hamilton TM glass syringe in a liquid displacement circuit. With volumes divided by 2, precision and repeatability are then much higher! Which diluents can I use? easyspiral Dilute works with saline solutions (e.g.: Ringer's solution, physiological saline). Avoid using diluents with nutrients as they may cause contaminations. Bacteria only remain a few seconds in the beakers before being deposited on the Petri dish so using a saline diluent has no effect on the fi nal enumeration. It is however possible to use nutrient solutions as diluent in the semi-automatic "My Diluent" mode. In that case, fi ll in the beakers with 4.5 ml diluent before processing. I need to make a 10-7 dilution. How can I do it? The easyspiral Dilute can do as many dilutions as you wish. Do a 10-5 dilution, take the last beaker and place it as the mother sample and then proceed with a new automatic dilution sequence. 6

Spiral plating method validation Purpose To study the reliability of Spiral plating with easyspiral comparing to the reference method (classical surface plating). Equipment and methods Five bacterial strains (three gram+ and two gram-) are tested: Lactobacillus casei var. rhamnosus, Bacillus cereus (ATCC 11778), Bacillus subtilis (ATCC 6633), Escherichia coli (ATCC 8739) and Pseudomonas aeruginosa (ATTC 9027). Bacterial cultures are diluted in buffered peptone water then plated on Petri dish agar (90 mm), using classical method and Spiral method (50!L in exponential mode). After 48h of incubation at 37 C (±1) on MRS (De Man, Rogosa, Sharpe) for Lactobacillus casei and 18h (±2) at 37C (±1) on PCA (Plate Count Agar) for Bacillus cereus, Bacillus subtilis, Pseudomonas aeruginosa and on Macconkey for Escherichia coli, counting is made with an automatic colony counter (Scan 1200). Results analysis Counting results are expressed in Log CFU/mL (Colony Forming Unit) and a regression analysis is made to assess the correlation between, Spiral method with easyspiral Pro and classical plating method.
