4 Warnings Before operating your FlowCam Macro analyzer, read this guide carefully to prevent damage to humans, animals, integrated devices, and connected devices. Always follow local safety rules. The instrument should be verified for safe operation following normal use, maintenance, and any factory service activities. Failure to use this instrument in the manner specified in this guide may impair safety features and nullify operator protection. Cautions To reduce the risk of electric shock, do not remove any protective covers. Safety Precautions To reduce the risk of serious personal injury or equipment damage, never remove the inner panels of the FlowCam Macro analyzer. Beware of electrical shock hazards. To prevent the risk of electrical shock or fire, do not locate the instrument in wet or unsafe laboratory conditions. Do not touch any damaged, non-isolated electrical parts. Use appropriate precautions to minimize and prevent the introduction of liquid into the instrument and contact with any electrical assemblies. The instrument operates on a detachable power supply that allows for immediate disconnect from power in the event of an emergency. Use only the power cords provided by the manufacturer to operate the instrument and peripheral devices. Mishandling may harm this instrument. Before lifting or moving the instrument, ensure that all cords and external equipment have been properly disconnected. When moving the instrument, always carry it with both hands. Grasp the front corner with one hand and place the other hand under the opposite side back corner for support. Always use proper lifting techniques. Install the instrument in such a way as to ensure that all power cords and ventilation openings are easily accessible and clear from obstruction with a minimum distance requirement of 10 cm. Do not operate or store this instrument outside of the following ranges: o o o Operation: 4 40 C, humidity 0 90% (noncondensing) Storage: C, humidity 0 95% (noncondensing) Maximum operating altitude: 2000 meters Do not attempt any repairs yourself. 5 Introduction The FlowCam Macro Dynamic Imaging Particle Analyzer uses proven dynamic imaging particle analysis (DIPA) to rapidly evaluate particulate matter in a moving fluid. Optimized for larger particles, the FlowCam Macro analyzer provides rapid particle characterization beyond just size. Direct, image-based measurements of particle size and shape enable differentiation of particle types in a heterogeneous mixture. 7 Overview The FlowCam Macro analyzer uses dynamic imaging particle analysis to perform particle measurements rapidly enough to produce statistically significant amounts of particle data. The FlowCam Macro analyzer gathers numerous measurements for each particle, providing the detailed data needed for a thorough particle analysis. A high-intensity LED light source combines synchronously with a very short shutter speed in the camera to freeze the moving particles in a fluid medium. The high-resolution camera, coupled with optics from the lens, captures thousands of particle images per second. As the camera captures each frame, the VisualSpreadsheet software extracts the particle images from the background in real time and stores them for analysis. The software then subjects each image to a series of more than 30 mathematical measurements to characterize that particular particle. Dynamic imaging particle analysis uses digital images to measure the size and shape of each particle. The sample containing the particles streams by the optics in the flow cell, and the camera captures thousands of particle images per second. FlowCam Macro Dynamic Imaging Particle Analyzer 7 of 34

8 Welcome This guide explains how to set up, operate and maintain your FlowCam Macro Dynamic Imaging Particle Analyzer. It also covers basic features of the VisualSpreadsheet software application that will assist you with the initial setup and operation of the analyzer. Package Contents Your shipping container includes the following items: FlowCam Macro analyzer with power cord Monitor with power cord and connectors USB keyboard USB mouse High-speed peristaltic pump system (optional) Accessory case with two flow cells (2 x 10 mm, 5 x 10 mm) Tubing and connectors Depending upon your specific analyzing needs, it may also include the following: FlowCam Macro sample mixing system FlowCam Macro sample collection container Ensure that there is no damage to the packing materials or equipment inside. If anything has been damaged or is missing, contact Fluid Imaging Technologies or your local distribution representative immediately. Be sure to save the white shipping container that your FlowCam Macro analyzer came in. Note: Use two people when lifting the analyzer. Depending upon its configuration, it could weigh in excess of 50 pounds. Choosing a Location Locate your FlowCam Macro analyzer in a well-ventilated area that is free of vibration and dramatic shifts in temperature. Place the analyzer on a clean, level, sturdy base capable of holding up to 100 pounds. Be sure to allow space for the monitor, keyboard and mouse, as well as the peristaltic pump and its components. For power, you need three outlets one each for the analyzer, the monitor, and the peristaltic pump. Use a stable power supply with surge protection and battery backup. Computer Peripherals Unpack the monitor, keyboard, mouse, and their accompanying cables and power cord. These peripheral components connect to the back of the FlowCam Macro analyzer, which contains the VisualSpreadsheet software program. 9 Setting Up the Analyzer Connecting Peripherals Connection ports are on the back of the FlowCam Macro analyzer. Two USB3 ports VGA port Two Ethernet ports Two RS-232 serial ports Audio in Audio out Microphone in HDM1 port DVI port Four USB2 ports 1. Connect the keyboard and mouse USB cables to two of the USB ports on the back on the analyzer. 2. Connect the VGA cable to the monitor s VGA port and the other end to the VGA port on the back of the analyzer. 3. Connect the monitor s power cord to the AC input port on the monitor and plug the other end into a power outlet. 4. Turn on the monitor s power switch. Powering on Your Analyzer 1. Connect the analyzer s power cord to the AC input port on the back of the analyzer and plug the other end into a power outlet. You will hear the analyzer s internal fan turn on. 2. Press the round power button on the top right of the analyzer to initialize the pump and the computer. The power button turns blue and you should see the startup process begin on the monitor. 3. After a few moments, the Microsoft Windows operating system opens and the icon for the VisualSpreadsheet software appears on the desktop. Installing the Flow Cell The flow cell is a high-precision optical component. Great care is taken during its manufacture to ensure conformance to exacting tolerances. The optical faces undergo dozens of inspections to ensure uniformity, from the certification of raw materials to the matching of the finished cells. The flow cell is fragile. Handle it carefully to ensure you do not damage it. Do not touch the faces of the flow cell with your fingers. Wear gloves when handling it. FlowCam Macro Dynamic Imaging Particle Analyzer 9 of 34

10 Misuse will gradually degrade the precision surfaces of the windows until significant reductions in transmittance occur due to light scattering. Improper cleaning or handling of cells can lead to degradation and may also compromise image quality and flow cell longevity. Cell windows must be clean and free of contaminants that could degrade performance. Effort is made to supply cells that are exceptionally clean. To keep them that way, be sure to clean the flow cells as described in the Care and Maintenance section. To install the flow cell: 1. Remove the flow cell from its protective case and inspect it for any damage. 2. Clean the outside of the flow cell with a piece of lens paper to remove any particles or fingerprints. 3. Connect the tubing from the sample inlet port to one end of the flow cell and the tubing from the sample outlet port to the other. 4. With the flow cell s frosted side toward you, place it into the analyzer, fitting it into the associated opening. 5. Place the applicable black retainer into the slot so that the flow cell is held in place and tighten the two thumbscrews. 11 Setting Up the Peristaltic Pump System The FlowCam Macro analyzer system includes an externally controlled high-speed peristaltic pump. The pump head accepts several different tubing sizes for a wide range of flow rates. The unique lever actuator design and automatic tubing retention allow quick tubing changes. This pump is designed to transfer or dispense liquids of various viscosities, as well as solids suspended in liquids. It is not intended to pump flammable or explosive substances. A B D A C A: Quick-release connectors B: Peristaltic pump C: Pump drive D: Optional stir-pak assembly Note: Be sure to read the user documentation that accompanies the peristaltic pump, and become familiar with its features and safety warnings before configuring it for use. To set up the pump: 1. Unpack the pump components, and set the pump drive on a dry flat surface next to the FlowCam Macro analyzer. 2. Connect the peristaltic pump to the pump drive and set it on a sturdy surface above the drive. 3. Connect the drive s power cord to the AC input port on the back of the drive and plug the other end into a power outlet. FlowCam Macro Dynamic Imaging Particle Analyzer 11 of 34

12 4. Connect one of the lengths of FlowCam Macro tubing to the FlowCam Macro analyzer s sample inlet port, and another length of tubing to the analyzer s outlet port. Note: The FlowCam Macro tubing includes quick-release connectors on one end that allow easy connections to the ports. Inlet/Outlet port with connector and tubing Inlet/Outlet port with connector and tubing 5. Load the tubing that is connected to the analyzer s sample inlet port into the pump head: a. Open the pump head by moving the actuator lever counterclockwise. b. Insert a loop of tubing into one open tubing retainer, between the occlusion bed and the rollers and into the other tubing retainer. c. Position the tubing so that it is firmly against the rollers and centered on the length of the roller. d. While holding the tubing ends, move the actuator lever back to the far clockwise (right) position. The pump will automatically grip the tubing. 13 6. Place the ends of the input and output tubing into the sample container. Note: The FlowCam Macro system can also be set up with a sample collection container. The sample collection container has a 300-µm sieve within the cover that can be used for zooplankton samples. The sieve retains any particles larger than 300 µm so they are not counted twice. Additional sieve sizes are available. The sample collection container can also be used without the sieve. Note: The VisualSpreadsheet program does not control the fluidics of the high-speed peristaltic pump, so you must enter its flow rate into the software s Context settings. Fluid Imaging Technologies recommends the flow rates below, but you should test your pump s output volume to determine the flow rate setting that is best for your operation. See Appendix A for more information. Actual flow rate through the FlowCam Macro analyzer is dependent upon the size of the flow cell, the diameter and type of tubing, sample viscosity, particulate loading, and the desired processing rate. However, the values below are good guidelines to follow to prevent double imaging and blurred images with a water-based solution. 14 Focusing the Flow Cell To focus the flow cell, you need the Fluid Imaging focusing standard, other bead standards, or the sample you will be analyzing. 1. On the monitor s desktop, double click the VisualSpreadsheet icon to open the program. 2. From the tool bar, click the Setup and Focus tool to open a live view of the camera window on the flow cell. The acceptable region lies within the red lines. 3. If necessary, align the flow cell to the correct position using the positioning knob located to the left of the flow cell assembly in the interior of the analyzer. Loosen the adjacent positioning lever to move the positioning knob. The flow cell can be moved left and right. FlowCam Macro Dynamic Imaging Particle Analyzer 14 of 34

15 Note: The red lines need to be positioned within the gray lines, which represent the walls of the flow cell. Align the red indicator lines as close as possible within the flow cell. 4. Introduce the sample through the peristaltic pump and stop the pump so that the sample is suspended in the flow cell. You can also run the pump at a slow speed (6 rpm). 5. As the sample flows, focus the view by turning the micrometer (inside the analyzer) clockwise or counter-clockwise until the edges of the particles become clear. Tip: Try to have a dense enough sample present in the camera view to ensure you are focusing the majority of the particles to a consistent optimum level, rather than just a few of them. 6. 16 Defining the Context Settings 1. On the monitor s desktop, double click the VisualSpreadsheet icon to open the program. 2. On the toolbar of the main page, click the Context tool. The Context dialog opens. The Context dialog contains the settings used by the FlowCam Macro analyzer to capture particles of interest during a sample analysis. This includes settings for the camera and other various parameters. The settings can be saved in a context file (filename.ctx) within a folder that contains all the files for a particular analysis. Basic software settings from the factory and your local default settings are also saved in context files. 3. Click the individual tabs (Load, Capture, Flow Cell, Fluidics, Filter, Stop) to enter the desired applicable settings. Note: See the VisualSpreadsheet documentation for additional information. 4. The Load tab lets you save your settings to a context file, or load a specific context file that you have previously saved. FlowCam Macro Dynamic Imaging Particle Analyzer 16 of 34

17 5. The Capture tab lets you set specific parameters for capturing particles, such as: Distance to Nearest Neighbor Any particles that are equal to or less than this distance in microns from each other will be imaged as one particle. Close Holes (iterations) Closing holes is a standard morphological operation used to fill in small gaps in an image that may have been lost due to a threshold setting being too high or the particle image being too faint. Collage Image Border Padding (pixels) Keeps an additional border of pixels around a particle when cropping from the raw camera image. Save Collage Images Saves all images (segmented particles from a raw camera image) captured during a run. Save Binary Collage Images Saves a visual overlay of the pixels determined to be a particle. Save Raw Camera Images Saves each raw camera image (that is, the image of the camera field of view) so that images can be reprocessed with different context settings. Max Raw Images Defines the maximum number of raw images to be saved when the Save Raw Camera Images option is selected. FlowCam Macro Dynamic Imaging Particle Analyzer 17 of 34

18 Particles Defined by Pixels These parameters provide threshold values for the software during routine acquisition. The density of the particle is defined relative to the background and the illumination. Acceptable Region These values define the region in an image where particles are captured. 6. The Flow Cell tab lets you select the type of flow cell being used, which automatically enters its depth and width. Select one of the macro options (FC2000x2mm or FC5000x5mm) from the drop-down list. 19 7. The Fluidics tab is where you define the sample volume, flow rate, autoimage rate, efficiency, run time, and sample dilution parameters. The Manual prime with sample option must be selected. Note: The flow rate must be set manually. See Appendix B for instructions about determining an appropriate flow rate. 8. The Filter tab lets you select an image filter (ESD, ABD, none, advanced) and set its minimum and maximum diameter values to specify the size range of the particles of interest to save. 9. The Reports tab provides options for exporting and printing reports when an analysis finishes, as well as options for performing classification. 20 10. The Notes tab is blank until you open a context file or list file. These files are stored as text files in the sample run data folder. 11. When an analysis finishes, the Run Summary tab displays summary information about the sample analysis. This data is saved as a text file in the sample run data folder. The filename begins with the List filename appended with _run_summary.txt. 12. The Stop tab offers options for terminating an analysis under certain conditions. You can select multiple options and the analysis stops when the first condition is met. Note: The operator can manually override any stop setting by closing VisualSpreadsheet. 21 13. When you have finished defining parameters, click OK. The analyzer is now ready to analyze samples. Performing a Sample Analysis Before Beginning Before running a sample analysis, ensure the following: You have installed a clean flow cell. Input and output tubing is connected to the analyzer securely. Input tubing is loaded in the external pump head. Input and output tubing is placed in the sample source. Powering on the System 1. Turn on the main power supply unit. 2. Press the round power button on the top right of the analyzer to initialize its internal pump and the computer. The power button turns blue and you should see the startup process begin on the monitor. 3. When the operating system finishes loading, the icon for the VisualSpreadsheet software appears on the desktop. 4. Double-click the VisualSpreadsheet desktop icon. Preparing the Software 1. Click the Context tool and define the Context Settings for the sample. 2. Click the Setup and Focus tool and focus the flow cell. FlowCam Macro Dynamic Imaging Particle Analyzer 21 of 34

22 Running the Sample 1. Turn on the power switch for the external peristaltic pump. 2. Prime and flush the tubing with ml of labgrade water. 3. Click the AutoImage tool on the VisualSpreadsheet toolbar. The Stop Conditions dialog opens. 4. Enter any desired notes and click OK. 5. Click Save to enter a filename and save your list file. The default filenaming scheme is the Julian day and time on the FlowCam Macro computer. 6. The analyzer begins pulling the sample, the AutoImage window indicates that the analyzer is calibrating, and the Collage window opens so you can see the images being captured. 7. When the sample finishes, the Collage window closes. Viewing Data 1. Click the Show All button on the toolbar to see the results. 2. Click the Open View button to see the images. Tip: You can stop the AutoImage process at any point. Shutting Down the System Rinsing the Analyzer Flush the system thoroughly after use to remove any leftover particles. Completely flush residual fluid from the tubing and flow cell. Rinse the flow cell after each use. Do not leave the flow cell with sample or material inside when not in use. Powering Down the Peristaltic Pump Turn off the power switch on the peristaltic pump. Disengage the tubing from the pump head and thoroughly rinse it with labgrade water after each use. Powering Down the Analyzer 1. Close the VisualSpreadsheet program. 2. Push the power switch on the top right of the analyzer cover. The computer shuts down and the monitor turns off automatically. Note: The FlowCam Macro analyzer s fluidics components continue to operate, along with the system s internal fans. Turning off the main power supply unit shuts down the entire system. 3. 23 Care and Maintenance General Immediately after use, rinse the analyzer system and tubing with appropriate cleaner. This rinse will help keep the flow cells clear and free from contamination for day-to-day use. If necessary, precede the rinse with a solvent or surfactant to remove particles. Flow Cell Immediately after use, or if you suspect that the flow cell is obstructed, remove the flow cell from the upper and lower tubing connections, and clean it as follows: o o o o o Use a warm water jet first, followed by dilute detergent, and then a warm water jet to rinse. Do not use an ultrasonic cleaner even cells that are fully thermally fused are at risk in ultrasonic cleaners. Do not blow dry cells with compressed air. Never allow cells with tap water on them to air dry. Use a final rinse of deionized water. Never leave a flow cell with sample or material inside when not using the analyzer. Inspect the flow cell channel frequently for contamination or obstructions. The inspection should include removing the tubing from each end to look into the channel. Clean the exterior of the flow cell before installation with deionized water and a low-lint lens paper. Analyzer Always disconnect the power cord from its power source before cleaning the analyzer. Clean the outside of the analyzer with a damp (not wet) lint-free cloth. A mild disinfectant or liquid soap will remove grease. Never blow canned or compressed air into the analyzer. Peristaltic Pump No lubrication is required for the pump head. Use a mild detergent solution or 70% isopropyl alcohol only to clean the pump head. Do not immerse the pump nor use excessive fluid. The pump requires no maintenance, other than cleaning. There are no user-serviceable or replaceable parts inside. Inspect tubing regularly and replace as needed. 24 Appendix A: Determining the Pump Flow Rate The VisualSpreadsheet program does not control the fluidics of the high-speed peristaltic pump, so you must enter its flow rate into the software s Context settings. Fluid Imaging Technologies recommends the flow rates below, but you should test your pump s output volume to determine the flow rate setting that is best for your operation. Recommended Flow Rates Actual flow rate through the FlowCam Macro analyzer is dependent upon the size of the flow cell, the diameter and type of tubing, sample viscosity, particulate loading, and the desired processing rate. Supplies Needed: 4-L beaker, 1-L graduated cylinder, water, tubing, pump, stir-pak and stand, pump controller, timer, data sheet To check the output volume: 1. Set your pump s rpm to a designated setting. 2. Flush the internal tubing line fully with water and ensure there are no air bubbles. 3. Start the pump flow and timer for 1 minute. 4. Measure the output volume (ml) in a graduated cylinder and record the data (use the data sheet on page 26). 5. Conduct three trial measurements for each pump setting. 6. In a graphical program, such as Microsoft Excel, use the equation below to graph rpm vs flow rate and draw a trend line: flow rate=slope*(rpm)-y-intercept 7. 27 Appendix B: Tubing and Quick-Release Connectors FlowCam Macro Tubing We recommend having a set of upper and lower tubing attached to the 5-mm flow cell, and another set attached to the 2-mm flow cell. Tubing is 1 cm in diameter. Upper tubing (6.5 cm long) Lower tubing (18.5 cm long) Replacing Tubing To remove the tubing from the analyzer sample inlet/outlet ports, loosen the bulkhead nut, pull the tubing from the bulkhead stem, and remove the nut from the tubing. When replacing the tubing, thread it through the bulkhead nut, ensure that it is seated firmly against the base of the bulkhead stem, and tighten the nut firmly. 30 Appendix D: Services Training Let our knowledgeable staff teach you how to operate and effectively use your FlowCam Macro analyzer and VisualSpreadsheet software. Hands-on training with a technical customer support technician provides the most thorough training experience because it is designed specifically for your application. Fluid Imaging Technologies offers four types of training/learning opportunities. Initial training usually occurs on-site at your group s facility soon after your FlowCam Macro analyzer arrives. This training is hands-on and suitable for a small group of 4 5 individuals. FlowCam Macro workshops are one- or two-day trainings or demonstrations held at Fluid Imaging Technologies headquarters in Scarborough, Maine. Our laboratory houses our legacy and our new 8000 series FlowCam analyzers. We can walk your group through our latest software and instrumentation using your own sample that you provide. Also, it s an opportunity to visit beautiful Maine! On-site refresher training is appropriate for on-boarding a new group of FlowCam Macro analyzer users, reviewing training topics for a new field season, or training existing employees who need a refresher. Remote learning sessions (RLS) are ideal for refresher training, for on-boarding new employees, or for our international customers whose travel expenses might be cost-prohibitive. RLS training occurs on your FlowCam Macro analyzer and VisualSpreadsheet program on a computer via Citrix Go to Assist as our secure method of screen sharing. The session can be recorded upon request. Preventive Maintenance Service A preventive maintenance service is designed to help ensure that your FlowCam Macro analyzer is always operating at its optimum. Preventive maintenance service includes: Certification that your FlowCam Macro analyzer is calibrated using NIST-traceable standards Thorough evaluation and cleaning of all accessory parts Testing of all hardware and software Replacement of common wear items, such as tubing and fittings (consumable items, such as flow cells, are not replaced) VisualSpreadsheet software upgrade for the FlowCam Macro analyzer and one satellite license A preventive maintenance service conducted at your site includes a half-day refresher training session with a Fluid Imaging Technologies technician. 31 Installation and Operational Qualification An installation and operational qualification (IOQ) service is performed when the instrument must be qualified for use. IOQs occur either during initial receipt of a FlowCam Macro analyzer or after it is relocated within a company property. IOQ service is only performed on-site at the customer facility. Documentation is completed, reviewed by a company representative, and signed. A certificate of completion is awarded, and a seal is adhered to the instrument upon successful completion. A field service report is completed and a copy of all documents are provided. Laboratory Services We offer a full suite of FlowCam models for particle analysis and method development services, customized to meet your needs. Particle analysis services Includes a guaranteed turnaround time of five business days. In consultation with your company, we run your samples, provide the associated data files using Box.com, and hold a follow-up seminar to discuss the results of your data. Method development services Offers a customized approach for sample optimization using your material in consultation with one of our laboratory technicians. The service includes determining proper sample dilution, system configuration, and standard operating procedures (SOPs) that you can use in your laboratory. 32 Standard Factory Limited Warranty Fluid Imaging Technologies, Inc. (the Seller ) warrants that the FlowCam Macro product (the Product ) purchased by you (the Customer ) shall be free from material defects in workmanship and material for a period of one year from the date of shipment by the Seller (the Limited Warranty ); provided, however, that the Limited Warranty does not cover any consumables (flow cells, flow cell holders, or tubing) or third-party manufactured/customer purchased items incorporated in the Product. For service and spare/replacement parts, the Seller also warrants the services it performs, and the spare and replacement parts it installs, for a period of one year from the date of performance of such services and the date of installation of the spare or replacement parts, respectively. The Customer understands and agrees that the Limited Warranty shall only apply if the Customer has used the Product in accordance with all specifications, documentation, and other information provided to the Customer by the Seller. The Customer s sole and exclusive remedy and the Seller s entire liability under the Limited Warranty shall be i) at the Seller s option, repair or replacement of the Product or any defective Product components (including any labor or other services related thereto), and ii) all shipping costs related to the repair or replacement of the Product or any defective Product components for both on-site and offsite repairs and replacements. Any Product or Product components returned to the Seller must have prior approval and must reference a return material authorization (RMA) number issued by the Seller. In the event the Seller determines that the entire Product must be returned to the Seller for repair or replacement, such Product must be shipped in its original shipping container to ensure adequate protection during transit. If the original shipping container is not available, a new shipping container may be purchased from Fluid Imaging Technologies at an additional cost. The foregoing Limited Warranty is in lieu of all other warranties, written or oral, express or implied, including, but not limited to, a warranty of merchantability, non-infringement, title or fitness for a particular purpose. In no event shall the Seller be liable for any direct, indirect, consequential, punitive, incidental or any other damages of any kind whatsoever, arising out of or relating to the Product, any Product components, any specifications, documentation or other information provided to the Customer in connection with the Product, or the Limited Warrant set forth herein, even if the Seller has been advised of, or otherwise should have been aware of, the possibility of such damages, and regardless of the legal theory or basis for such claim. (the Seller ) will repair or, at its option, replace, without charge, your FOV flow cell (the Product ) if it proves to be defective in material or workmanship under normal use during the warranty period. This Limited Warranty does not apply to damages or functional issues that result from misuse, abuse, use outside environmental specifications, improper installation, unauthorized modification or alteration, and acts of God.
