POLYCHROME DEVICES (MONOCHROMATOR) Polychrome Devices (Moochromator) Overview This module allows you to cotrol aalogue devices, such as the TILL Photoics Rapid Moochromator, remotely from your computer. It allows you to move to a differet wavelegth istataeously usig the slider or butto cotrols o the Polychrome Palette. You ca also store up to four wavelegths betwee 260 m ad 680 m ad move to them istataeously by just clickig o the store ico. I order to use these devices, you eed: A Multifuctio PCI card (1200) 1 A BNC breakout box A BNC lik cable The appropriate Opelab Hardware Support module The Opelab Polychrome module No additioal driver software is required for the card. The Polychrome ad Hardware Support modules are ot part of the core product, but ca be purchased separately. Refer to Chapter 2 of the Opelab Core Guide for istructios o istallig modules. Oce you istall the Polychrome module, you will see a ew palette o the scree. This palette cotais all the cotrols for the aalogue device. You may also otice a additioal software item whe you load Opelab, for example the TILL Hardware Support module. This software cotrols commuicatio betwee the computer ad the hardware cotroller. Refer to the maufacturer s documetatio for iformatio o settig up ad coectig devices. Refer to the documetatio accompayig the board, ad to your Macitosh maual, for iformatio o istallig the board. 1 You ca also use a Digital I/O card, but you will eed additioal driver software, which is istalled i the System folder i the Extesios folder. POLYCHROME DEVICES (MONOCHROMATOR) The Polychrome Palette Oce you have istalled the Polychrome module, the Polychrome Palette will be available wheever you ru Opelab. Click to display the Calibratio dialogue Move slider to far left to tur off device (resets to 680 m) Use the buttos to fie tue the wavelegth i 1 m icremets Use the slider cotrol to select the wavelegth (coarse cotrol) Click o the store ico to select the wavelegth store Calibratig the TILL Polychrome II Each Polychrome II is supplied with: Two small filters (380 ad 505 m wavelegths) A photodiode filter holder A voltmeter You must calibrate agaist two wavelegths. The calibratio will ot apply util you have etered the calibratio for the secod wavelegth. You must also remember to Polychrome 3
OPENLAB use the Reset butto i Opelab s Polychrome Calibratio dialogue to remove ay previous settigs before you eter your ew calibratio settigs. The followig istructios take you step by step through the process. To prepare for calibratio 1. Plug the red jack plug from the photodiode ito the ohm-milli amp socket, ad the black jack plug ito the COM socket of the voltmeter. 2. Switch o the lamp ad wait for at least 20 miutes. 3. Remove the optical fibre cable from the microscope codeser. 4. Switch the mai dial of the voltmeter to 2000m, which should give a low readig. 5. Put the two filters ito the filter sockets i the photodiode (mirrored surfaces outwards), ad rotate the holder util the blue filter (380 m) is i frot. 6. Positio the optical fibre so that it illumiates the 380 filter ad select 380 as a positio o the Polychrome Palette. The voltmeter will show a readig. To calibrate the Polychrome 1. Start the calibratio process by displayig the Polychrome Palette ad clickig o the Calib. Butto. 2. Click o Reset to remove ay previous settigs. 3. Click o OK ad select 380 o the Polychrome Palette. Use the small left/right arrows to adjust the value o the Palette i steps of 1 m. Cotiue util you get the maximum readig o the voltmeter. 4 Opelab v. 2.0.7

Whe you have achieved the highest voltage readig you ca get, click o the Calib. butto ad type i 380 to replace the value that is registerig o the Polychrome Palette. For example, the Palette may thik you are at 385 m, whe you are actually at 380 m. 5. Now positio the 505 m filter i frot of the photodiode (the 505 has the silver surface). 6. Select 505 o the Palette ad repeat the process described i steps 3 ad 4 (but type 505 i the Calibratio dialogue). 7. Click o OK. As log as you have etered the values for both positios, the calibratio ca adjust the scale o the Palette to match the device. The Polychrome is ow calibrated ad ready for use. Settig Up Wavelegth Stores If you are workig maually, the wavelegth stores allow you to assig values for up to four chaels so that you ca quickly select the required wavelegth. (I a automatio you ca just use the Set Wavelegth task to set a wavelegth ad move to it. See page 8.) 1. Click o oe of the umbered store icos to select it. You will otice a black outlie aroud the store ico o the Palette. 2. Use the slider cotrol ad buttos to assig a wavelegth to the store. Note the color bar uder the slider cotrol, ad how the color of the ico reflects the selected wavelegth. 3. Repeat steps 1 ad 2, selectig store icos ad assigig wavelegths to them for the other three stores, as appropriate. Polychrome 5
OPENLAB Usig the Palette Oce you have set up a store, just click o it to move the device to that wavelegth. To edit a store, just click o it ad use the slider cotrol ad buttos agai to assig a ew wavelegth to the store. If you wat to tur off the sigal, move the slider cotrol to the far left. This resets the wavelegth to 680 m. Note: I maual operatio, you ca oly switch to the Off positio usig the slider cotrol. 6 Opelab v. 2.0.7
POLYCHROME DEVICES (MONOCHROMATOR) Polychrome ad Automator tasks If you have the Automator module, you will be able to use the Polychrome tasks i your automatio. The task will appear i the Task List i the Automator Widow. This sectio describes the Polychrome task. Refer to the Automator Referece Maual for further details about creatig ad ruig automatios. OPENLAB Select Store Task This task allows you to select oe of the pre-stored wavelegths. You must set up the wavelegth store (as described o page 5) before you ru the automatio. Use this task if you eed to kow that a particular store i your automatio always has a certai value. For example, you could use it to set up the Off positio of 680m. Eter the id of the wavelegth store Set Wavelegth Task This task allows you to assig a wavelegth value to oe of the four wavelegth stores ad move to it. Oce you eter the value, it is assiged to the store. I this way, you ca vary the wavelegth of the store as, ad whe, required i your automatio. POLYCHROME DEVICES (MONOCHROMATOR) A Example Automatio I the followig automatio we will capture a umber of images at two wavelegths. The Target image widow task targets the image widow for the captured images. The Loop task sets up the loop for the umber of images that you ited to capture. The first Set Wavelegth task moves the Polychrome II to the 340 m positio. The Capture Layer task captures the layer at 340 m. The secod Set Wavelegth task moves the Polychrome II to the 380 m positio. The Capture Layer task captures the layer at 380 m. Note: If you use this routie i a automatio, remember to build i time for the user to mout the ext slide. (Use a Tell task or a Delay task.) Polychrome 9
