PRECAUTIONS The Microwave Fixation System is developed to rapidly halt brain chemical activity prior to the analyses of neurotransmitters or protein phosphorylation in the brain of the small rodents such as rats and mice. If the apparatus is used for a purpose different from what it originally intended, Muromachi will not be responsible for any damage that operators suffer. Further, it is designed for use by qualified personnel only. Be sure to read and understand the entire Instruction and Operation Manuals before attempting to operate the Microwave Fixation System. All users must be aware of and pay specific attention to the following: Only a qualified electrician should remove the Microwave Fixation System s cover plate. Likewise, system servicing and/or maintenance must only be performed by a qualified electrician. The Microwave Fixation System is a high voltage device. During operation, a voltage of approximately 5,000V is present within the device. To prevent electrical shock turn off the main and outside power switches before removing the system cover plate for servicing. There are a number of small holes around the applicator head that allow the animal to be viewed within the fixation system. While microwave energy will not leak from these holes one should never insert metal objects (or similar items) into these holes. Doing so may result in electrical shock and/or damage to the unit. Do not splash water on the device. Water on the unit may result in electrical shock or charring of the device. The Microwave Fixation System is not designed to be dust or moisture resistant. Special attention should be paid to the area surrounding the system. Keep this area clean and dry to ensure that the unit functions properly. Do not block the unit s airflow. The airflow inlet and outlet are located at the front and back of the instrument respectively. Appropriate airflow is required for proper functioning of the system. The Microwave Fixation System is equipped with caster wheels that aid in positioning of the unit. Lock casters in place once the location and position of the instrument is set. Locking the casters will prevent the unit from unexpected moving during operations.

1. INTRODUCTION MMW-05 is a system composed of a microwave power generator (SM1150) and a specially designed applicator unit that radiates a large amount of microwave energy in a short period of time on a rat or mouse restrained in the holder inside the applicator head. Exposure duration can be varied from 0.1 sec. to 2.99 sec. by means of a digital timer. Power level is also variable 2 to 5 kw max. 2. Pre-heating time required: For optimum stability 30 min warm-up is recommended. Operators should stay at a distance of 35cm away from the applicator head just in case. 3. 4. SYSTEM COMPONENTS 4-1 Power Supply The power supply unit provides various sources of electric power necessary to operate the magnetron, and consists of the high-tension source of 3-phase full wave rectified negative voltage to the magnetron cathode, the magnetron filament power source, the magnetron exciting power source, the relays to turn on and off the various sources, circuit breakers to protect each circuit when any abnormal condition takes place, and fuses. 4-2 Touch Panel Display Push buttons on the display provide various controls necessary to the system operation. Exposure timer ranges from 0.1 sec. to 2.99sec. by 0.01 sec. steps. Anode current is variable from approximately 0.6A to 1.5A (max.), or 2kW to 5kW in microwave output power level. 4-3 Magnetron The source of microwave energy employed is the Panasonic Magnetron 2M265-M11. Using direct heating cathode, 2M265-M11 requires a shorter preheating time, also has a higher conversion efficiency to microwave power of 70% (nominal). Its air-cooled anode operates at 5 kv. 4-4 Three-Stub Tuner This tuner allows the microwave power to reach inside the applicator efficiently. The tuner is already factory-adjusted to the matching condition for each of the applicators to be used. It should be noted that an arbitrary resetting could cause an inefficient transmission of the microwave power. 4-5 Applicator Head The unit accommodates an animal holder, containing a rat or mouse, onto which microwave power is radiated. 4-6 Safety Circuit For the sake of system safety and protection, detecting circuit is provided as described below. It is interlocked to prevent starting or to stop the microwave irradiation if any abnormality is found before or during operation. - 3 -

6. OPERATING PROCEDURE 1. Supply power to the system and turn the MAIN switch clockwise to the ON position till it clicks. The following message will appear on the Touch Panel screen. Fig 1 NOTE: When the Lock Lever of the MAIN switch is open (Fig A), the MAIN switch will not be turned. Push in the Lever (Fig B) to turn the switch to the ON position (Fig C). Fig A Fig B Fig C 2. GREEN light indicates Normal status RED light indicates Abnormal 3-1. When RED light(s) is on, the MAIN Switch (NFB) may be turned off. Do whatever action is necessary until all the GREEN lights come on. 3-2. When all the GREEN lights come on, the display will move to the WARM-UP screen below. * Cooling fan starts rotating * WARM-UP countdown starts and min to go will be displayed. Fig 3 For optimum stability 30 min warm-up is required. 3-3. When the 30 min warm-up is over, the following menu will appear on the screen and the word READY will appear. Fig 4-6 -

Push the CHANGE button to enter the output power level and exposure time desired, respectively. If you want to change the numeric values entered, push the CLR key and input numeric values desired. Push the ENTER button to determine the input. b. If the numeric values entered are out of acceptable range, the word OVERFLOW appears for approximately 2 seconds. - 7 -

4. Push the OK button to move to the next step. The following message will appear on the screen. Fig 6 5. If the current settings are all right, place a loaded restrainer into the applicator head and push the OK button. The following menu will appear on the screen. Fig 7 NOTE: Push the BACK button to go back to the previous screen (Fig 4) to change the parameters. 6. Make sure that a loaded restrainer is placed in the applicator head. WARNING: NEVER OPERATE MICROWAVE FIXATION SYSTEM WITHOUT A LOADED RESTRAINER INSIDE. MUROMACHI WILL NOT BE RESPONSIBLE FOR ANY DAMAGE CAUSED BY THIS. - 8 -

7. Push the START button twice within 0.5 second to execute microwave irradiation. The word EXPOSURE appears and flashes on and off during irradiation. Fig 8 8. After the successful exposure period is over, the EXPOSURE disappears and the word FINISHED appears. Fig 9 If you push the STOP button before irradiation, you can go back to the previous screen (Fig 6). If the STOP button is pushed during exposure, irradiation will cease and the screen will move to the Fig 9 (FINISHED). - 9 -

9. Take the restrainer out of the applicator. The display will show as below. The machine becomes available for next operation after the 3-min pausing time ( recovery time ). Fig 10 The display will also go back to the same screen (Fig 10) by pushing the OK button. 10. If performing multiple irradiations or multiple experiments, repeat from step 4 to step 9. 11. When all experiments are finished, 11-1. Push the EXIT button. The following message will appear on the screen. Fig 11-10 -

11-2. If you push the BACK button, you can go back to the Fig 10 and continue operation. 11-3. If you push the EXIT button here, 20 min COOL-DOWN will start. Fig 12 11-4. 7. TROUBLESHOOTING In the event of trouble, high voltage will be cut off to stop irradiation and red light indicating the trouble will come on. 8. HOW TO USE WATER-JACKETED ANIMAL HOLDER 8-1 Filling up animal holder with water a) Fill up water through the water inlet (the right side tubing) using a disposable syringe. CAUTION: Fill it with water gently so that water gets into the cone as uniformly as possible. Big bubbles should be removed, however, you don t have to be very concerned. b) When filling is completed, put the male lure fitting on the other female lure fitting before pulling out the syringe tip so that water may not flow out. c) Pull the syringe out of the right tubing. Now the water-jacketed animal holder is ready for use. The male lure fitting put on the left side tubing should be kept attached till the irradiation is over. 8-2 Water replacement Water becomes hot after irradiation and therefore, should be replaced with new water every time a new animal is used. - 13 -

9. EXPOSURE CONDITIONS 9-1 Exposure Conditions of Mouse Brain Applicator Head: TAW-174P + WJM-28 Water-jacketed Mouse Holder BODY WEIGHT OUTPUT POWER (kw) EXPOSURE TIME (sec) 20-30g 5.00 kw Approx 0.94 sec BEST HOLDER POSITION NOTE: 43mm Uniform heating can be obtained with the mouse holder at the position shown in the above photo. If the output power is insufficient, less uniform heating will be obtained. In such a case, increase the exposure time in 0.1 sec steps. If the holder is positioned at the place other than the above, it will be difficult to get a uniform heating. The positioning of the restrainer is critical to the repeatability. The above exposure conditions are just for your reference. Output power varies with each magnetron which is the power source of the system. Therefore, every user is requested to develop his or her own optimum exposure settings empirically. - 14 -

Otherwise it will be difficult to get uniform heating because animals try to move backward when radiated. The positioning of the animal is critical to the repeatability. a) Insert the animal holder into the applicator head till it stops at dead end. For repeatability, it is also important that the restrainer be positioned in the same place each time. * If you find front part of the rat brain not heated well, adjust the holder position a little backward. b) The above exposure conditions are just for your reference. Output power varies with each magnetron which is the power source of the system. Therefore, every user is requested to develop his or her own optimum exposure settings empirically. - 15 -

10. APPLICATOR HEAD REPLACEMENT PROCEDURE When replacing applicator heads, make sure that the Main Switch is in the OFF position.
