Image Itesifier Image Itesifier Overview This module provides support for the Hamamatsu M4314 Image Itesifier cotroller. This is a separate device, which is coected via a serial lik to oe of the serial ports o the Macitosh. Oce you have coected the Image Itesifier to the serial port, you eed to tell the software about it. You use the Prefereces dialogue to do this. The Image Itesifier is ofte used with the Argus-20 computer. The diagram o the followig page illustrates the cablig betwee the various uits; the camera head, the Image Itesifier, the Macitosh ad the Argus-20. OPENLAB Image Itesifier Cablig The Video coectio from the Argus-20 to the Macitosh may be to either the built-i video port or to a separate card i the Macitosh. IMPORTANT! Make sure that the Argus-20 ad the Macitosh are switched off before you attach the SCSI cables, ad always switch o the Argus-20 computer before you switch o the Macitosh. Image Itesifier Serial Prefereces Before you ca start to use the Image Itesifier with Opelab, you eed to tell Opelab which serial port the image itesifier is coected to. 1. 2. Select Prefereces... from the Edit meu. Click o the Serial Ports ico i the left-had colum. 3. Set up the Image Itesifier o the appropriate port. (See Chapter 18 of the Opelab Core Guide for further details about coectig serial devices.) 4. Click o the Save butto. 5. Quit ad relauch Opelab. Image Itesifier 3

Image Itesifier Usig Opelab to Cotrol the Image Itesifier This sectio describes how to use the Image Itesifier palette to cotrol the Hamamatsu device. If you are usig the Image Itesifier with a Argus-20 computer, refer also to the Argus-20 User Guide. 1. Click o the ico to switch o the high voltage supply. You will otice a flashig warig symbol o the palette. 2. Use the Sesitivity slider cotrol to adjust the amplitude of the sigal or check the Automatic Sesitivity box (so that there is a X i it) to allow the Image Itesifier to adjust the gai automatically. 3. Eable, or disable, ay of the other check box cotrols, as appropriate. Note: If you wat to cotrol the Gai maually, you must first disable Automatic Gai. Image Itesifier 5

Image Itesifier Set Sesitivity This task allows you to set the sesitivity. Either eter a value for the sesitivity or click o the check box to use Automatic Sesitivity. Either eter a expressio to set the sesitivity value Or check to use automatic sesitivity Set High Voltage This task allows you to tur the high voltage o or off. Click o the appropriate butto. Image Itesifier A Example Automatio The followig automatio uses a Argus-20 ad a Image Itesifier to expose a sample to light for five secods, before photo coutig the results. Settig up the automatio The automatio is set up as follows: The Target Image Widow task defies the target image widow i Opelab o the Macitosh. Make sure that you select the empty image documet o which you have already defied a ROI set to the area that you wat to sca for photo levels. The Stop Imagig task esures that the Argus-20 is idle before photo coutig starts. Image Itesifier 9
OPENLAB The Set PC discrimiator level task sets the level at which evets will be couted. The Photo Coutig task clears the memory ad specifies the cout period. The Loop task uses the cout period value to cotrol how may cycles the tasks withi the loop will complete. The Set Illumiatio task switches the light o, ad the Wait task esures that the sample is exposed for five secods before aythig further happes. The Set Sesitivity ad Set High Voltage tasks set the maximum gai ad switch the high voltage o, respectively. The Start Imagig task starts the photo coutig process. The Get Imagig Status task checks whether the photo coutig process is curretly active, ad puts the retured status value ito a variable called imagestatus. The If task the checks the imagestatus value. If it is 1, the the photo coutig process is still active, ad the automatio will cotiue checkig the status util it returs a value of 0, which idicates that the photo coutig process has fiished. The Set High Voltage task switches the high voltage off. The Trasfer Image task trasfers the processed image to the Macitosh as a ew layer.. 10 Image Itesifier
